The Canine Distemper Virus-Parvovirus-Dirofilaria immitis Ag, Ehrlichia canis-Leptospira spp. Ab Combo Test is a lateral flow immunochromatographic assay designed for shelter and rescue health screening by detecting canine distemper virus antigen in secretions from the eye conjunctiva, nasal cavities or anus, canine parvovirus antigen in feces, and Dirofilaria immitis antigen together with antibodies to Ehrlichia canis and Leptospira spp. in serum, plasma or whole blood, from a single test card.

Detection Principle

The test is based on sandwich method lateral flow immunochromatographic assay. The test card features 5 testing windows, each containing a T (test) zone and a C (control) zone that are initially invisible before the assay. When the treated sample is applied, the liquid flows laterally across the NC membrane of each test strip, interacting with pre-coated target-specific reagents respectively. The presence of target analytes in the sample results in a visible T line, while the appearance of the C line indicates a successful assay.

Test Procedure

1	Equilibrate — allow all components to equilibrate to room temperature (15–25 °C) for 10 minutes before use.
2	Collect samples — collect secretions from the eye conjunctiva, nasal cavities or anus with a swab for the CDV window and collect fresh feces with a separate swab (covering about 1/3 to 2/3 of the swab tip) for the CPV window; collect whole blood into a vacuum blood collection tube or obtain serum or plasma by centrifugation for the CHW, EHR and LEPTO windows.
3	Prepare samples — insert the CDV swab into the CDV assay buffer tube and the CPV swab into the CPV assay buffer tube, agitate each swab in its matching buffer, and use the liquid supernatant as the treated sample for the CDV and CPV windows; serum, plasma or whole blood is used directly without dilution.
4	Apply samples — remove the test card from the foil pouch and place it flat on a work surface. Transfer 3 drops of the treated CDV sample into the CDV sample well and 4 drops of the treated CPV sample into the CPV sample well; for each of the CHW, EHR and LEPTO sample wells, add 1 drop of serum, plasma or whole blood with a capillary dropper, then 3 drops of the CHW-EHR-LEPTO assay buffer with a disposable dropper.
5	Read result — start the timer and read the result at 5–10 minutes. Do not move the test card during testing.

The five windows are read independently, and any faintly visible T line should be considered positive when the C line is also present. The CDV window detects canine distemper virus antigen in secretions from the eye conjunctiva, nasal cavities or anus; the CPV window detects canine parvovirus antigen in feces; the CHW, EHR and LEPTO windows detect Dirofilaria immitis antigen and antibodies to Ehrlichia canis and Leptospira spp., respectively, in serum, plasma or whole blood. Read the result at 5–10 minutes after sample application; a window in which no line appears in the C zone is invalid.
