About Canine Relaxin

Canine relaxin is a peptide hormone primarily produced by the placenta during pregnancy in dogs, with smaller amounts secreted by the corpus luteum in the ovaries. Belonging to the insulin-like hormone family, its key role is to prepare the reproductive tract for pregnancy and facilitate parturition (birth). It helps relax the pelvic ligaments and soften the cervix to accommodate fetal growth and ease delivery. Relaxin levels rise during early pregnancy, peak in mid-to-late gestation, and decline after whelping. It is commonly used as a diagnostic marker for pregnancy in dogs, detectable via blood tests as early as 20–25 days post-mating.

 

Clinical Signs

Relaxin itself does not cause disease, but its presence or fluctuations are associated with physiological changes during pregnancy:

  • Pregnancy-Related Changes: Enlarged abdomen due to fetal growth, mammary gland development (swelling, nipple enlargement), and behavioral shifts (e.g., nesting behavior, increased affection or restfulness).​
  • Abnormal Scenarios: In cases of false pregnancy (pseudopregnancy), mild relaxin elevation may occur, leading to similar signs like mammary enlargement or nesting, though no fetuses are present.
     

Canivet Relaxin Test

Relaxin is a specific hormone generated mainly by the placenta. It is usually used as an indicator of pregnancy status in female animals. In pregnant animals, relaxin can be detected from 15 days after the nidation of fertilized egg in the uterus wall. The RLN Test is a rapid test to detect relaxin in animals that may be pregnant. The suggested detection period is 28-35 days after mating. Too early detection may lead to a negative result when relaxin in serum or plasma is too low. It is suggested to repeat the test after 1 week if a negative result was observed.
