Detection Principle

The test is based on a competitive lateral flow immunochromatographic assay. The test card features a testing window with an initially invisible T (test) zone and a C (control) zone. When the treated sample is applied, the liquid flows laterally across the NC membrane; the target analyte in the sample competes with the labeled analyte component on the test zone for binding to the limited gold-conjugated antibody sites, so the signal intensity of the T line correlates with the analyte concentration in the sample. The optoelectronic system of the VETSEE G2 Dry Immunoassay Analyzer analyzes the signal intensity of the T line against the lot-specific calibration curve (imported by scanning the QR code), and the concentration is displayed directly on the analyzer screen. The C line indicates a valid test result.

Test Procedure

1	Equilibrate — allow all components to equilibrate to room temperature (15–25 °C) for 10 minutes before use.
2	Turn on the analyzer — power on the dry immunoassay analyzer and use its scanner to scan the QR code on the inner side of the box lid or the bag label to import the calibration curve of this lot.
3	Load the test card — remove a Cortisol test card from the foil pouch, position it front side up with the sample well oriented towards the outside of the test card slot, and insert the card into the slot.
4	Prepare sample — use a pipette to add 100 μL of assay buffer into a centrifuge tube, then add 100 μL of serum or plasma specimen into the same tube and mix thoroughly to obtain the treated sample.
5	Apply sample — use a pipette to add 100 μL of the treated sample into the sample well and push the test card to the farthest end of the slot.
6	Run the test — tap [STANDARD TEST] on the screen; the cortisol concentration will be displayed automatically at the end of the countdown, about 10 minutes later.

The cortisol concentration is displayed directly on the screen of the analyzer (detection range 0.5-30 μg/dL); refer to the reference range in practice. When the analyzer displays "invalid", it is recommended to review the operating procedures and perform another test. Reference interpretation is provided in the context of adrenal function testing: for the ACTH stimulation test, administer ACTH at 2.2 IU/kg IM (maximum 40 IU) and measure cortisol after 2 hours; for the low-dose dexamethasone suppression test (LDDST) give 0.01 mg/kg dexamethasone, and for the high-dose dexamethasone suppression test (HDDST) give 0.1 mg/kg dexamethasone, measuring cortisol at 4 and 8 hours.
