Product Overview
The Feline Cortisol Test is a lateral flow immunochromatographic assay designed for the quantitative detection of the concentration of cortisol in feline serum or plasma, providing a numerical result in about 10 minutes to help monitor adrenal function in cats during ACTH stimulation or dexamethasone suppression testing.
Suitable for:
🐈 Cats at risk of infection 🏥 Veterinary clinics & animal hospitals 🐾 Rescue centers & shelters 🔬 Field screening

Detection Principle
The test is based on a competitive lateral flow immunochromatographic assay. The test card features a testing window with an initially invisible T (test) zone and a C (control) zone. When the treated sample is applied, the liquid flows laterally across the NC membrane; the target analyte in the sample competes with the labeled analyte component on the test zone for binding to the limited gold-conjugated antibody sites, so the signal intensity of the T line correlates with the analyte concentration in the sample. The optoelectronic system of the VETSEE G2 Dry Immunoassay Analyzer analyzes the signal intensity of the T line against the lot-specific calibration curve (imported by scanning the QR code), and the concentration is displayed directly on the analyzer screen. The C line indicates a valid test result.
Test Procedure
| 1 | Equilibrate — Allow all test components to equilibrate to room temperature (15-25°C) for 10 minutes before use. |
| 2 | Turn on the analyzer — Turn on the dry immunoassay analyzer and use its scanner to scan the QR code on the inner label of the box or the label of the bag to import the calibration curve of the lot. |
| 3 | Load the test card — Remove the test card from the foil pouch. With the front facing up and the sample well oriented towards the outside of the analyzer's test card slot, insert the card into the slot. |
| 4 | Prepare the sample — Use a pipette to add 100 μL of assay buffer into a centrifuge tube, then add 100 μL of the specimen into the same tube and mix thoroughly; use the mixed liquid as the treated sample. |
| 5 | Apply the sample — Use a pipette to aspirate the treated sample and add 100 μL of it into the sample well, then push the test card to the farthest end of the test card slot. |
| 6 | Run the test — Tap [STANDARD TEST] on the analyzer screen; the result will be displayed in about 10 minutes. |
* The result is calculated and displayed by the VETSEE G2 Dry Immunoassay Analyzer (Standard Test); refer to Result Interpretation for the reference range.
Result Interpretation
| Concentration | Interpretation |
|---|---|
| ACTH stimulation test, post-ACTH (2 h): <2 μg/dL (<55 nmol/L) | Consistent with hypoadrenocorticism; treatment with mineralocorticoid and/or glucocorticoid |
| ACTH stimulation test, post-ACTH (2 h): 2-6 μg/dL (55-166 nmol/L) | Inconclusive |
| ACTH stimulation test, post-ACTH (2 h): 6-18 μg/dL (166-500 nmol/L) | Normal |
| ACTH stimulation test, post-ACTH (2 h): 18-24 μg/dL (500-663 nmol/L) | Equivocal, possibility of Cushing's |
| ACTH stimulation test, post-ACTH (2 h): >24 μg/dL (>663 nmol/L) | Consistent with Cushing's; perform a high-dose dexamethasone suppression test to discriminate between ATH and PDH |
| LDDST (0.01 mg/kg dexamethasone), cortisol at 4 h or 8 h: <1 μg/dL (<28 nmol/L) | Normal |
| LDDST (0.01 mg/kg dexamethasone), cortisol at 4 h or 8 h: 1-1.4 μg/dL (28-39 nmol/L) | Inconclusive, repeat in 6-8 weeks |
| LDDST (0.01 mg/kg dexamethasone), cortisol at 4 h and 8 h: >1.4 μg/dL and >50% of baseline | Consistent with Cushing's; perform a high-dose dexamethasone suppression test to discriminate between ATH and PDH |
| LDDST (0.01 mg/kg dexamethasone), cortisol at 4 h or 8 h: >1.4 μg/dL and <50% of baseline | Consistent with PDH |
| HDDST (0.1 mg/kg dexamethasone), cortisol suppressed at 4 h or 8 h: <1.4 μg/dL or <50% of baseline | Consistent with PDH |
| HDDST (0.1 mg/kg dexamethasone), cortisol not suppressed at 4 h and 8 h: >1.4 μg/dL and >50% of baseline | Run further testing to differentiate PDH from ATH; consider measuring ACTH and/or performing an abdominal ultrasound |
The concentration of cortisol is displayed directly on the analyzer screen (detection range 0.5-30 μg/dL). Cortisol levels are interpreted in the context of the dynamic protocol performed: the ACTH stimulation test (ACTH 2.2 IU/kg IM, maximum 40 IU, cortisol measured after 2 hours), the low-dose dexamethasone suppression test (0.01 mg/kg dexamethasone, cortisol measured at 4 and 8 hours) or the high-dose dexamethasone suppression test (0.1 mg/kg dexamethasone, cortisol measured at 4 and 8 hours); refer to the reference rows above. When the screen displays "invalid", review the operating procedures and repeat the test with a new card. The suggested reference range was established based on a certain region; it is recommended that each laboratory establishes its own reference range to fit local patients.
* Suggested reference ranges were established based on a certain region; because of varietal and regional differences, each laboratory should establish its own reference range. For in vitro veterinary use only — results should be considered alongside other clinical information.

FAQ
| Q | Which specimens can be tested? |
| A | Feline serum or plasma. |
| Q | How long until the result? |
| A | About 10 minutes after starting the test on the analyzer. |
| Q | Do I need an instrument? |
| A | Yes — the VETSEE G2 Dry Immunoassay Analyzer is required; the concentration is calculated and displayed automatically. |
| Q | How is the concentration reported? |
| A | The analyzer displays the concentration directly on its screen (detection range 0.5-30 μg/dL); compare it with the reference range in Result Interpretation. |
| Q | Can the test card be reused? |
| A | No. Each card is for single use only. |
| Q | What does the C line mean? |
| A | The C (control) line must always appear after sample application, indicating proper test performance. No C line = invalid test, repeat. |
